This should be old news for even the n00biest of n00bs, but sometimes it bears repeating.
Test, test, and test.
Case in point. I started a simple video blog about 2 months ago. I chose a theme, put some posts up, added viScript, dumped some Adsense blocks on it and forgot about it. About a week ago, I noticed that it had jumped to #7 for it's primary keyword and started looking at the Adsense and traffic. Traffic has climbed to about 200 - 250 uniques per day and my Adsense was making a whopping $0.30 to $0.50 per day. Not too great, but not bad for ignoring something for so long.
So, I decided it was time to revamp the site. I purchased a domain for it (previously it was a subdomain of one of my test sites), changed the theme and started messing with Adsense blocks. Oh yeah, and I redirected the old subdomain to the new domain.
Anyhow, I saw an immediate jump in CTR and revenue. CTR jumped from about 1% to 5% and revenue has gone up to as much as $3 per day. Two days ago, I removed an adblock to see if that would push the CPC up for the link units (they were getting more clicks), but amazingly it didn't, it actually dropped for the link units. So yesterday I re-added the adblock that I had removed, but when I re-added it, I took the border off of it. Weird thing happed, my CTR went up even more, and my link units CPC went back to what it was before.
So, the moral to the story is Always Be Testing. Try new themes, ad placement, different colors, different adblocks, etc.
Oh yeah, and another cool thing, since changing to the new domain, I'm now ranked in the top 5 for my primary keyword and getting about 120 - 150 visits per day from Google alone. Not bad for a site that started out as a joke.
