Well read up on the pareto principle. (wikipedia link)

Namely, 20% of the work will turn in 80% of the profits.
In web sites, these magical 20% are normally divided by content/usability/readability.

Concentrate on those points.
Then do design, then do minor fixes, then go pixelpushing, etc...

Don't make the user think! Get him where he wants to be easily.

That's the way that site makes money.
That's the way ebay makes money.
That's why amazon has 1-click checkout.
That's...etc, etc.... you get it, I am sure.
